Decreases in the function of γ-aminobutyric acid, dopamine, serotonin, and opioid peptides, as well as dysregulation of brain stress systems such as corticotropin-releasing factor and neuropeptide Y are hypothesized to contribute to a shift in reward set point. It is hypothesized further that the pathology of this neurocircuitry is the basis for the emotional dysfunction long associated with drug addiction and alcoholism in humans. From the perspective of a given drug, it is unknown whether the hypothesized reward dysfunction is specific to that drug, common to all addictions, or a combination of both perspectives.
